Description

SUMMARY, EXPLANATION AND LIMITATIONS:
Cadherins are a family of multifunctional calcium-dependent transmembrane glycoproteins, which promote and maintain the cellular adhesion in practically all multicellular organisms. Cadherins superfamily consists of more than forty proteins, with a homology between them of 50-60%. Cadherin expression is required for the assembly of cells in solid tissues. Cellular interactions are due to homophilic interaction between extracellular regions of molecules similar to cadherin present in neighbor cells. Recent studies of crystallographic analysis of a cadherin extracellular domain suggest that cadherin molecules cooperate between them for form a continuous line of cellular adhesion. In binding zones, cadherins anchor to actin cytoskeleton through small cytoplasmic proteins, like for example β-catenin and g-catenin, which bind to α-catenin (actin binding protein). Cadherins, besides they play an important role in differentiation process and tissue morphogenesis, they also play a significant role in the tumorous invasion modulation and metastasis. Its expression is inversely correlated with the invasive capacity of tumorous cells. Besides, the tissue specificity of the different cadherin types makes them important markers for identifying and differential diagnoses of certain tumor types.

Clone: 36B5
Isotype: IgG1
Immunogen: Recombinant prokaryotic protein representing the amino-terminal external portion of the E- cadherin molecule.
Staining pattern: Cell membrane.
Positive control: Tissue sample from tonsil.

APPLICATIONS:
This antibody is designed for the specific localization of human E-cadherin using IHC techniques in formalin-fixed, paraffin-embedded tissue sections.
It is recommended for evaluating the E-cadherin expression in primary cancers and metastatic lesions; and for distinguishing between ductal mammary carcinomas, where is positive, and lobular carcinoma, where normally is not detected.
